Insight into Ntrust Clinical Trials
The Ntrust program comprises two primary clinical trials: Ntrust-1 (NCT06557265) and Ntrust-2 (NCT06733935). Both trials are open-label and multi-center, focusing on dose-escalation in patients diagnosed with autoimmune diseases. Participants will undergo lymphodepletion followed by receiving CD19-targeted CAR-NK cell therapy. These trials primarily aim to assess the safety of NKX019 and its potential to induce lasting remission by resetting the immune system through the removal of harmful B cells.
The Ntrust trials plan to enroll up to 12 patients per dose level for each disease being studied. Key indications include myositis and systemic sclerosis, alongside other conditions such as ANCA-associated vasculitis, rheumatoid arthritis, lupus nephritis, and primary membranous nephropathy. If necessary, additional patient enrollments may occur to better define participant populations for future studies.
Throughout the studies, participants will receive a cycle of three doses of NKX019 following a lymphodepletion regimen using fludarabine and cyclophosphamide, or cyclophosphamide alone if they exhibit considerable cytopenia. This strategic framework aims to evaluate the action of NKX019 as a single agent and to expedite the journey toward regulatory approval. Participants in both trials also have the option to receive further cycles if necessary to enhance or deepen their response.
About NKX019
NKX019 is a cryopreserved, off-the-shelf immunotherapy that utilizes NK cells extracted from the blood of healthy adult donors. Engineered with a humanized CD19-directed chimeric antigen receptor (CAR), NKX019 aims to improve cell targeting. A proprietary membrane-bound form of interleukin-15 (IL-15) enhances its persistence and action without the dependence on additional cytokine support. CD19 serves as a marker for both healthy B cells and those involved in autoimmune disorders. Nkarta is actively investigating NKX019's efficacy across several autoimmune conditions.
About Nkarta
Nkarta is at the forefront of developing allogeneic, readily available NK cell therapies aimed at assisting those affected by autoimmune diseases. The company seeks to leverage the innate immune system's capabilities to eradicate pathogenic B cells that trigger autoimmune responses. Nkarta's innovative CAR-NK cell therapies offer quick access and outpatient administration, making these treatments accessible to a wider range of patients experiencing various autoimmune conditions.
