GPN Vaccines Secures $10 Million to Advance Revolutionary Pneumococcal Vaccine

GPN Vaccines has successfully closed a $10 million financing round to further develop Gamma-PN™, a novel serotype-independent pneumonia vaccine.

GPN Vaccines, Inc. has successfully finalized a private placement financing round, raising US$10 million. This funding aims to propel the company's lead vaccine candidate, Gamma-PN™, into the next phase of clinical development.

Funding Objectives

The proceeds from this investment will be directed towards several critical initiatives. These include the manufacture of cGMP batches, enhancing the company's laboratory capabilities for opsonophagocytic activity (OPA) testing, and preparing to submit an Investigational New Drug (IND) application to the U.S. Food and Drug Administration (FDA).

Innovative Design of Gamma-PN

Gamma-PN is an investigational pneumococcal vaccine designed to address the shortcomings of existing serotype-specific vaccines. Currently, over 100 serotypes of Streptococcus pneumoniae exist, yet available vaccines offer protection against only a limited number. This leads to an ongoing need for manufacturers to develop new vaccines that include additional serotypes.

What sets Gamma-PN apart is its targeted approach to generate immune responses against protein antigens shared across all pneumococcal strains. This method is expected to provide a layer of protection regardless of the capsular serotype.

Expert Insights

Tim Hirst, Ph.D., Chairman and CEO of GPN Vaccines, commented on the innovative nature of Gamma-PN. He stated, “For decades, pneumococcal vaccine development has concentrated on increasing the number of serotypes included in complex formulations. Gamma-PN seeks to break away from this trend by focusing on underlying protein antigens common to all strains, aiming for broad protection independent of serotypes.”

Clinical Development Progress

Gamma-PN has already undergone four Phase 1 clinical trials, demonstrating notable safety and immunogenicity across diverse adult demographics. Preliminary data also show that the vaccine stimulates functional OPA antibodies capable of bacterial killing across various serotypes. GPN is now gearing up for a multinational, randomized, double-blinded, controlled study to further investigate the vaccine's safety, tolerability, and immunogenicity in adults aged 50 and above.

Market Potential

The pneumococcal vaccine sector represents a significant market, currently valued at over $9 billion annually. Leading products include Pfizer's Prevnar® and Merck's Vaxneuvance® and Capvaxive®. However, the market faces challenges due to the limitations of existing serotype-specific vaccines, which complicate manufacturing processes and supply chain logistics.

Bruce Forrest, M.D., Ph.D., MBA, Executive Chief Scientific Officer of GPN Vaccines, expressed optimism about Gamma-PN's potential impact. “The size of the pneumococcal vaccine market highlights the urgent need for effective vaccination. Traditional vaccines target individual serotypes, but Gamma-PN opens doors to generating broad immunity across multiple strains without the need for inclusion of each serotype in the formulation,” he explained, emphasizing its capacity to disrupt the market's current paradigm.

Future Developments

Gamma-PN™ is GPN Vaccines' flagship investigational pneumococcal vaccine, crafted to offer extensive, serotype-independent defense against Streptococcus pneumoniae. Its unique formulation aims to elicit immune responses against commonly shared protein antigens, potentially protecting against over 100 known serotypes as well as future strains. Following four successful Phase 1 studies, GPN is preparing to advance Gamma-PN into Phase 2 clinical trials.

Beyond Gamma-PN, GPN Vaccines is working on a broader portfolio of bacterial vaccines, including the Gamma-SU program targeting Streptococcus suis and additional programs focused on non-typeable Haemophilus influenzae and Moraxella catarrhalis.
